Inhibitory Effect of Euonymus alatus on the Chicken Gamma Globulin- and Platelet Activating Factor-Induced Anaphylaxis in Mice

Abstract
This study was carried out to evaluate the effect of Euonymus alatus(EA) on the chicken gamma globulin(CGG)-induced active systemic anaphylaxis (ASA) and platelet-activating factor (PAF)-induced lethal anaphylaxis(LA) in ICR mice, and
degranulation
and
histamine release of rat peritoneal mast cells. Mice were sensitization i.p. with 500ug CGG plus Bodetella pertussis (10E6 cells) and 1mg alum on day 0, and were challenged i.v. with 500ug CGG on day 18 of sensitization. A severe ASA was induced
in
all
EA-untreated mice. However, the ASA-reaction was reduced when mice were given i.p. EA-water extract for 4 days before sensitization or challenge, and this reaction was completely abolished when mice were treated with EA for 4 days before
CGG-sensitization and 18 days after CGG, suggesting that EA has an active principles to prevent ASA. Intravenation of PAF(1 ug/mouse) alone produced a severe LA. But, the LA was not evoked when PAF was injected into mice in combination with
EA(0.5ug/mouse), or when PAF was injected into mice pretreated with EA(0.5ug/mouse, i.p) 30 min before PAF.
These results indicate that EA shows strong antagonism against PAF. And in vitro EA also remarkably prevented the degranulation of compound 48/80-treated mast cells, and reduced significantly the histamine-release of these cells. Taken together,
the
present study suggests that EA can be developed as a new drug for preventing and curing allergic diseases.
